I thought the title was meant to indicate that "Book of Eli" was a new book of the bible.
I also thought it obvious that his actions/abilities were the result of divine guidance.
His insistence that noone else touch the book (namely when Mila's character tried to take it from his bag to read it while he seemed to be sleeping) made me think it was in braille.
