Anti-colonialism, in this context, means that America shouldn't really be at the top of the tree, telling other countries what to do.

I don't find it a very convincing argument. Obama is a soft power president who wants to pursue America's foreign policy goals through diplomacy and talking, but he is still pursuing America's foreign policy goals, rather than some anti-colonial agenda.
I haven't seen the movie (and I don't think I'll bother) but I have read D'Souza's article about how Obama thinks. It pretty much tries to paint him as some outsider who doesn't think the way most Americans do - and that's why he wants to increase taxes on the rich, have universal healthcare coverage, etc - as though these ideas are un-American and that only a foreigner steeped in anti colonialism would seriously suggest them. But it's not as though any of these things are seriously outside the mainstream of American politics.
