He was dying of that curse and wanted Snape to kill him to entirely entrench Snape's identity as a loyal Death Eater. That's why Dumbledore was saying, "Please". Snape did not want to do it but he had promised to when the time came.
At the time Snape diagnosed the curse, he said that Dumbledore had around a year to live; that was the previous summer.
