You will have to wait until after your first week not working because the claim is retrospective (i.e. did you work the previous week?). There may also be a waiting week for benefits, though I believe most state have now waived that so you now get paid from week 1. Once a claim is established, it stays active for a year, so even when you go back to work, you can reopen the claim after any week of not working.
