Manual does cut down shifting time. You should try it. I can rank around top 1000 worldwide with manual but would be top 500 if I used manual+clutch.

Upshifting on manual+clutch saves about .6 seconds, and with about 10 shifts in a 1.5 minute lap that can be a full second difference in lap time.
Since there are players faster than I am using manual on the leaderboards, I haven't tried to get used to manual+clutch because I figure better to improve my skills at operating the car before worrying about operating the controller. Until I get better, I'm fine knowing there are a bunch of people ahead of me on the leaderboards only because of their shifting setting.
The real key is learning the weight shift in the car - how much time you lose braking too late for a corner and waiting for the car to lay back again so you can get on the throttle as soon as possible without going wide or spinning the tires. Tanner Foust had a good tip during a Forza Racing Championship broadcast: "When exiting a turn you should already be pointed directly and the next turn entry."
