In Windows 7, the Search box in the top-right of your Explorer window will search not only the filenames, but also the contents of a file. A Flash drive is not indexed by Windows by default, but that just means it'll take longer. I suspect that Google Desktop will also do it. And there's a hard drive searching tool called "Everything" (linked below) that I think has become moot since Windows 7 pretty much does the same thing.
